SeQuant® HILIC (hydrophilic interaction liquid chromatography) HPLC columns constitute a range of high-performance tools for separating polar hydrophilic compounds. All columns carry densely bonded, truly zwitterionic functional groups with a charge balance of 1:1. Separation is achieved by hydrophilic partitioning combined with weak ionic interactions for maximum selectivity, high loadability and easy optimization of methods. The SeQuant® HILIC family includes the ZIC®-HILIC, ZIC®-pHILIC and the ZIC®-cHILIC.

ZIC®-HILIC columns have documented excellent selectivity for separations of polar hydrophilic compounds in HPLC and LC-MS methods. These columns carry densely bonded, truly zwitterionic functional groups with a charge balance of 1:1. Separation is achieved by hydrophilic partitioning combined with weak ionic interactions resulting in maximum selectivity, high loadability and easy optimization of methods. The comprehensive ZIC®-HILIC product range includes classical analytical and capillary columns, as well as semi-preparative formats. All columns are available with 3 or 5 µm particles, and pore sizes of 100 or 200 Å.

ZIC®-pHILIC columns are the preferred choice when extreme eluent conditions are needed to overcome challenging separations of polar hydrophilic compounds. Offering excellent pH stability from pH 2 to 10, these polymer-based columns carry densely bonded, truly zwitterionic functional groups with a charge balance of 1:1. This results in straightforward and reproducible separations with wide-spectrum pH tuning for optimal separation selectivity and detection sensitivity. Furthermore, SeQuant® ZIC®-pHILIC columns minimize background noise in evaporative detection techniques with high sensitivity to ions, such as charged aerosol detectors (CAD). SeQuant® ZIC®-pHILIC columns are available with 5 µm polymer particles in various formats of analytical dimensions.

ZIC®-cHILIC columns provide complementary selectivity to optimize separations of polar hydrophilic compounds. The columns carry a novel densely bonded, 1:1 charge-balanced phosphorylcholine functional group. As with all true zwitterionic stationary phases, straightforward and reproducible separations are achieved by hydrophilic partitioning combined with weak ionic interactions.

The more accessible positive charges of ZIC®-cHILIC columns enable selectivity tuning in chromatography through altered interaction with anionic and cationic molecular moieties. This can be favorable in several situations, for example in the separation of labile metal complexes or highly polar and multiply charged aminoglycosides.

What is HILIC?

HILIC or hydrophilic interaction liquid chromatography is a high-performance liquid chromatographic (HPLC) technique for separation of polar and hydrophilic compounds. Simply put, HILIC is a normal-phase type of separation but uses reversed-phase type eluents. Thus, it unites the benefits of aqueous mobile phases with separation based on normal phase principles in which more polar compounds respond with higher retention. This results in more efficient separation. Furthermore, HILIC is easy to combine with a wide range of detection methods, and boasts improved sensitivity with some of these techniques, such as electrospray ionization mass spectrometry (ESI-MS).
